production and characterization of a murine monoclonal antibody against human ferritin
Authors
abstract
background: ferritin is an iron storage protein, which plays a key role in iron metabolism. measurement of ferritin level in serum is one of the most useful indicators of iron status and also a sensitive measurement of iron deficiency. monoclonal antibodies may be useful as a tool in various aspects of ferritin investigations. in this paper, the production of a murine monoclonal antibody (mab) against human ferritin was reported. methods: balb/c mice were immunized with purified human ferritin and splenocytes of hyper immunized mice were fused with sp2/0 myeloma cells. after four times of cloning by limiting dilution, a positive hybridoma (clone: 2f9-c9) was selected by elisa using human ferritin. anti-ferritin mab was purified from culture supernatants by affinity chromatography. results: determination of the antibody affinity for ferritin by elisa revealed a relatively high affinity (2.34×109 m-1) and the isotype was determined to be igg2a. the anti-ferritin mab 2f9-c9 reacted with 79.4% of hela cells in flow cytometry. the antibody detected a band of 20 kda in k562 cells, murine and human liver lysates, purified ferritin in western blot and also ferritin in human serum. conclusion: this mab can specifically recognize ferritin and may serve as a component of ferritin diagnostic kit if other requirements of the kit are met.

full textproduction of monoclonal antibody against human immunoglobulin
immunoglobulin e is one of the five classes of immonoglobulins that plays an important role in allergic diseases. production of monoclonal antibodies by a single clonotype against different epitopes of immunoglobulin e has high priority in development of diagnostic kits. in this study, an attempt was made to produce monoclonal antibodies against human immunoglobulin e. balb/c mice were immunize...
full textproduction of monoclonal antibody against human nestin
we have employed a peptide-based antibody generation protocol for producing antibody against human nestin. using a 12-mer synthetic peptide from repetitive region of human nestin protein devoid of any n- or o-glyco-sylation sequences, we generated a mouse monoclonal antibody capable of recognizing human, mouse, bovine, and rat nestin. a wide variety of nestin proteins ranging from 140-250 kda w...
full textProduction and Characterization of Monoclonal Antibody against Saffron Pollen Profilin, Cro s 2
Background: Allergy to Saffron (Crocus sativus) pollen has been described in people involved in processing of saffron flower stamens. Profilins have been identified as a pan-allergen in different plant pollens and foods. This molecule is an actin-binding pro-tein with a molecular weight of 12-16 kDa found in eukaryotic species. Objective: The aim of this study was to generate monoclonal antibod...
full textproduction and characterization of new anti-human cd20 monoclonal antibody
the b-cell cd20 antigen is one of the most reliable surface targets in immunotherapy of b lymphoma. in this project, we studied the production and characterization of a new monoclonal antibody against chimeric human cd20 extra loops (hcd20 exl). the results showed that clone c12h, igg2/k isotype reacted with the antigen in elisa and immunoblot. the kd value was found to be 2×10-9m and flow cyt...
full textTHE PRODUCTION OF MURINE MONOCLONAL ANTIBODIES (MAb) DIRECTED AGAINST HUMAN T- LYMPHOCYTE SUBSETS
The production of murine monoclonal antibody (MAb) has not yet been reported in Iran. The present work describes for the first time the generation of several murine hybridoma clones secreting MAbs directed against human leukocyte surface antigens. The secreted antibodies by hybridoma clones have been screened on different lymphoid and non-lymphoid tissues. Results indicated that of seven h...
